Favor specificity
In running your flow cytometry experiments, you will often want to boost the visibility of dull antigens to overcome possible interference from others. CytoGenie helps you do this whenever it can. Click Favor specificity.
A window will appear. Click on a reagent in the first column. Reagents that may limit its visibility will appear in the third column. Click on a grayed-out determinant to explain why it is not available.
A screen will pop up to explain.
Click on one of the available determinants whose potential interference you wish to subordinate to some degree. Then right-click or click the Downward options arrow.
CytoGenie will find color and laser combinations that make the highlighted determinant in the first column more visible than the one in the second column. Repeat the steps with other reagents in the first column that you wish to favor. When you have finished, click OK.
CytoGenie can sometimes minimize potential interference in a different way, which is generally less effective than favoring specificity. If you’d like to try it, right-click or click the Downward options arrow.
An Options pop-up menu will appear. Click Sort/arrange columns.
CytoGenie will display the Sort/arrange columns window, which determines the look of the Stain sets grid. In the example we’ve been using, click to hide the two URL columns and unhide the Color assigned and Handle assigned columns. Leave the Sort? and Sort order settings as they are. Click Done.
The Stain sets grid will now display the new columns. The green borders around two of the cells warn you of potential interference. Click one of them. Then click the Downward options arrow.
An Options pop-up menu will appear. Click Minimize interference.
CytoGenie may then ask you to confirm that you want to limit the list. For now, click Yes.
A list will appear. If all the entries are grayed out, Click Cancel.
Click on the other green-bordered cell. Then right-click or click the Downward options arrow.
On the Options pop-up menu, click Minimize interference.
On the confirmation window, click Yes.
A list will appear. If any of the entries are not grayed out, click the one whose interference you wish to minimize. Click Preview to see whatever details you have previously entered. Click Pick.
Repeat as you think necessary to remove other possible interference.
